Ochronne działanie ekstraktu rumianku zwyczajnego przed padaczką wywołaną PTZ: Wpływ na zapalenie mózgu, aktywację komórek pomocniczych i białka nerwowe
Protective effects of Matricaria chamomilla L. extract against PTZ-induced epilepsy: Targeting neuroinflammations, glial reactivity and neurofilaments

OBJECTIVES: The current study investigated the potential neuroprotective effect of Matricaria chamomilla L. extract (MCE) against PTZ-induced seizures in rats as well as its effects on the electrophysiological changes, neuroinflammatory cytokines, microglia and neurofilaments expression in hippocampus. METHODS: 36 male rats were randomly allocated into 3 equal groups, a) sham group, b) PTZ group (50 mg/kg i.p in 0.2 mL on alternate days for 2 weeks) and c) MCE group (200 mg/Kg MCE orally in 1.0 mL saline daily for 2 weeks with PTZ injection). Racine score and latency to the first sign of seizure were recorded with each PTZ injection. By the end of experiment, electrophysiological recording from CA1 region was done as well as biochemical assessment of oxidative stress markers (MDA, GSH and catalase), expression of IL6 and TNFalpha at level of mRNA and immunohistochemical assessment of Lc3, Nfl, NFH, IBA1 and GFAP were done. RESULTS: MCE considerably reduced the Racine score and lengthened the latency to the first seizure compared to PTZ group. Also, we observed a significant increase in delta and theta waves with a significant decrease in gamma waves in MCE group compared to other groups. When comparing the MCE group to the PTZ, we observed a substantial drop in MDA and an increase in GSH and catalase (p < 0.001), as well as a decrease in IL6 and TNFalpha expression at the mRNA level (p < 0.01). In addition, the MCE group showed a significant rise in NFL and Synaptophysin expression and decrease in IBA1, GFAP expression compared to the PTZ group (p < 0.001). CONCLUSIONS: In rats, MCE demonstrated neuroprotective and antiseizure effects against seizures brought on by PTZ. These effects may be associated with normalization of hippocampal electrical activity, reduction of oxidative stress and inflammatory markers, attenuation of glial activation, and restoration of NFL expression.
